Principle

Principle

The test is based on the IFCC kinetic method. ALT catalyzes the reaction between L-alanine and α-ketoglutarate, producing pyruvate and L-glutamate. The pyruvate then reacts with NADH in the presence of lactate dehydrogenase (LDH), causing oxidation of NADH to NAD⁺. The decrease in absorbance measured photometrically is proportional to the SGPT activity in the sample.
